Application of Flow-Injection Spectrophotometry to Pharmaceutical and Biomedical Analyses
The focus of this chapter is on methods in which we measure a time-dependent signal. Chemical kinetic methods and radiochemical methods are two examples. In this section we consider the technique of flow injection analysis in which we inject the sample into a flowing carrier stream that gives rise to a transient signal at the detector. Because the shape of this transient signal depends on the physical and chemical kinetic processes occurring in the carrier stream during the time between injection and detection, we include flow injection analysis in this chapter. Flow injection analysis FIA was developed in the mids as a highly efficient technique for the automated analyses of samples. FIA is one example of a continuous-flow analyzer, in which we sequentially introduce samples at regular intervals into a liquid carrier stream that transports them to the detector.
Various parts of the system such as pump, valve, and reactor may be built or adapted from available materials. Therefore the systems can be at lower cost as compared to other instrumentation-based analysis systems. Their applications for determination of biomarkers for liver diseases have been demonstrated in various formats of operation but only a few and limited types of biomarkers have been used as model analytes. Liver diseases hepatic diseases cover the broad range of liver disorders. The liver has many important functions including breaking down toxic substances in the body and excreting them into urine, producing and secreting bile to aid in food digestion, converting glucose into stored glycogen and vice versa , metabolizing ingested medicines to obtain active ingredients, and producing blood-clotting factors, amino acids and cholesterol to transport fat. Damage to the liver will cause disruption of these functions which can cause serious damage to the body. Even though the liver is considered the only organ in the body that has exceptional capability in replacing damaged cells, if too many liver cells are damaged, the liver may fail to perform properly.
The concept of flow injection analysis FIA was introduced in the mid-seventies. It was preceded by the success of segmented flow analysis, mainly in clinical and environmental analysis. This advance, as well as the development of continuous monitors for process control and environmental monitors, ensured the success of the FIA methodology. As an exceptionally effective means of mechanization for various procedures of wet chemical analysis, the FIA methodology, in use with a whole arsenal of detection methods of modern analytical chemistry, proved to be of great interest to many. The fast and intensive development of the FIA methodology was due to several factors essential for routine analytical determinations, such as very limited sample consumption, the short analysis time based on a transient signal measurement in a flow-through detector and an on-line carrying out difficult operations of separation, preconcentration or physicochemical conversion of analytes into detectable species. Twenty-year studies by numerous research groups all over the world have provided significant progress in the theoretical description of dispersion phenomena in FIA and various operations of physicochemical treatment of the analyte.

Flow injection analysis
Flow injection analysis FIA is an approach to chemical analysis. It is accomplished by injecting a plug of sample into a flowing carrier stream. FIA is an automated method of chemical analysis in which a sample is injected into a flowing carrier solution that mixes with reagents before reaching a detector. Over past 30 years, FIA techniques developed into a wide array of applications using spectrophotometry , fluorescence spectroscopy , atomic absorption spectroscopy , mass spectrometry , and other methods of instrumental analysis for detection. Automated sample processing, high repeatability, adaptability to micro-miniaturization, containment of chemicals, waste reduction, and reagent economy in a system that operates at microliter levels are all valuable assets that contribute to the application of flow injection to real-world assays.

Flow-injection spectrophotometric determination of tetracycline and doxycycline in pharmaceutical formulations using chloramine-T as oxidizing agent. Weinert; Helena R. Keywords: tetracyclines; flow-injection; spectrophotometry. Tetracyclines TCs represent a class of antibacterial compounds with broad spectrum antibiotics for their high activity against nearly all gram-positive and gram-negative bacteria.
